Evaluation of new plant type (NPT) lines of rice (Oryza sativa L.) for callus induction and in vitro plant regeneration

Abstract: An investigation was made to evaluate the callus induction and regeneration capabilities of nine new plant type (tropical japonica) lines compared to a high yielding indica rice variety, MDU 5, as control. The results of the experiment inferred that the combination of growth regulators containing MS + CH (1gl-1) + Kinetin (0.5 ml-1) + 2,4-D (2 mgl-1) was outstanding in promoting early callus induction, high callus induction percentage and fresh weight of callus. In the case of regeneration, the combination of hormones, namely, MS + IAA (3 mgl-1) + BA (3 mgl-1) + kinetin (1 mgl-1) was found to be the best in regenerating maximum number of plantlets. The new plant type line of rice, IR 75282-10-3-3-2, was identified for its better ability in callus induction and calluses with increased fresh weight and also the capacity to regenerate more number of plantlets. The findings of this experiment will be of much use in any future study, aiming to exploit the tissue culturing potentials of new plant type lines of rice.
